When children are aggressive, their voices lower and they sound angry. When they are playing, their voices are higher pitched, often laughing. If they are taking on a role, the voice may be lower, but in an exaggerated, almost singsongy way to indicate that they are playing. I find that teachers who allow for roughhousing can tell the difference fairly easily.
